Traditional tissue analysis typically uses frozen or paraffin-embedded sections. Micron-thick tissue sections enable the labeling and characterization of individual cells but do not allow the detection of three-dimensional (3D) features of biological samples. Section analysis can only detect and study a portion of a pathological sample, leaving a significant amount of information in the entire pathological tissue untapped. With the development of tissue transparency techniques and the birth of light sheet microscopy, researchers now can perform complete 3D imaging characterization of intact samples.

High-throughput sectioning and optical imaging analysis of tissue samples using traditional tissue analysis techniques are expensive and difficult to obtain 3D information. CD Biosciences can assist researchers using confocal microscopy or light sheet imaging to perform 3D imaging of tissue samples processed with CLARITY technology. Its state-of-the-art laboratory equipment and techniques allow scientists to perform 3D imaging and phenotyping of cell populations throughout the depth of the tissue for biological research.

For 3D tissue imaging, CD Biosciences scientists use CLARITY technology to clean tissues without the need for serial sectioning. This technology preserves spatial relationships and microstructure by forming a more permeable tissue-hydrogel mixture for deep optical inspection, which can be used for 3D mapping and identification of rare markers. In addition, the team analyzes the results using specialized image analysis software to obtain reliable results.

CD Biosciences has developed an optimized antibody labeling method that allows researchers to achieve uniform labeling on thick tissue while maintaining histological integrity. Upon request, CD Biosciences’ research team can use immersion objectives, light sheet microscopes and other imaging methods to perform 3D image analysis on larger tissues. In addition, CD Biosciences offers 3D image analysis services, although some customers only require raw data files from its imaging services.
